FIELD OF INTEREST
核内受容体は、発生・分化といった生理的因子あるいは環境因子等の種々の刺激に応答してリガンド依存的に遺伝子発現を調節する転写制御因子です。これら核内受容体は、糖尿病や動脈硬化症といった代謝異常症、薬物相互作用あるいは癌細胞の増殖にも関与しており、薬物開発のターゲットとして注目されています。私どもは核内受容体の生理的機能、組織あるいは細胞特異的な発現、リガンド特異的に形成される転写複合体を明らかにするため、モノクローナル抗体を作成して局在解析、ChIP-seq法、およびtargetedプロテオミクスを推進しています。
1)核内受容体の局在解析:タンパク質の組織や細胞局在に関するデータはそれらの生理的機能を類推する上で有用な情報源です。私どもは免疫組織学的検討によって核内受容体の組織および細胞局在について検討を行っています。
2)ChIP-seq法を用いた核内受容体結合部位の同定:ChIP-seq法はゲノムDNAと標的タンパク質との相互作用を免疫沈降法と数百万におよぶDNA断片を同時シークエンスすることが可能な高速シークエンサーとの組み合わせで特異的に検出する方法で、この手法を用いてゲノム上の核内受容体結合部位の同定を行っています。
3)核内受容体のtargetedプロテオミクス:タンパク-タンパク相互作用は核内受容体の転写活性およびその標的遺伝子の発現を規定しています。私どもは、ショットガンプロテオミクス法にて核内受容体タンパク複合体を同定しております。
