Expression of Pparg and Cebpa genes, master regulators of white adipocytes differentiation, is regulated by epigenome. The H3K4/H3K9me3 bivalent chromatin domains on Pparg and Cebpa genes function as a gatekeeper of adipocytes differentiation to integrally regulate the expression of genes involved in lipid accumulation (Mol Cell 2015). By targeting the chromatin domains, we are applying genome editing technology to epigenome editing to reveal epigenetic regulation of lipid accumulation. We aim to develop therapeutic agents for lifestyle-related disease targeing histone modifications.
